I live in Scarborough and own a two story semi detached house. Would like to know if I can extend by first floor by 15 feet in my back yard my backyard is 100 feet long.

It depends on a number of factors, size of the house (total livable area), local zoning bylaws and local regulations if near conservation area. The best first course of action would be to call the city and ask. If there are other houses in the neighborhood who have completed such an addition, then there is a good chance you can to.

Might be a bit tight. A survey wpuld have to be done, drawings/blueprints and apply for permit.
I did an addition in Ajax and we were allied to extend 12 ft.

I meant, in Ajax ,we were allowed 12 ft.
It is based on the size of the home against remaining backyard space.
We submitted a complete survey of the property ,together with updated blueprints of the entire home including the new proposed addition.

If your development regulations are similar to ours your restrictions will include a maximum percentage of lot coverage (in our case 28% for the home and 12% for the garage - totaling 40), and there'll be restrictions on the maximum distance from the front property line. Of course there may be many other restrictions/regulations.
If you're planning to build closer than 1.2 m to your neighbor you'll need to provide a fire resistance rating to the outside wall and of course no windows or doors.
